Necropsy

Necropsy

High quality histopathology starts with proper necropsy techniques.  HSRL has a team of trained prosectors who travel the country providing prosector services to you and your team.  

We also offer necropsy supervision by one of our highly trained veterinary pathologists.  Our team is trained in necropsy techniques on the following species:  mouse, rat, ferret, guinea pig, pig, rabbit, dog and non-human primate.

Histology

Histology

Having a well trained team of histotechnicians is important in preparing the highest quality slides.  HSRL employs a technical trainer who is a highly skilled histotechnologist and oversees the training of all laboratory personel.

Our team routinely prepares upwards of 15,000 prepared slides each month- which requires a large dedicated and skilled team of histotechnicians.  Each tissue leaving HSRL has been reviewed microscopically to ensure it is consistently placed, free of artifacts, and has crisp staining.

We offer: bone decalcification, oversized tissue paraffin embedding and processing, hundreds of special stains and work with a variety of species to include both small and large animals.

Our histology laboratory is 100% GLP compliant and has been audited by the FDA three times and Form 483s have never been issued.

Located in the heart of the biomedical research community in Worcester, MA, HSRL now operates a second GLP-compliant laboratory location dedicated to specialty histopathology and immunohistochemistry services. HSRL is prepared to support both GLP and non-GLP studies for all services offered in its VA and MA sites. Available lines of service at the MA facility include: immunohistochemical staining (IHC), immunofluorescent staining (IF), whole slide scanning and morphometric analyses.

IHC

IHC (GLP and non-GLP)

HSRL’s experienced team is proud to offer a broad range of routine and advanced IHC and IF staining techniques for both frozen and formalin-fixed, paraffin-embedded (FFPE) tissues. Numerous routine antibodies across multiple species have been validated in-house, including mouse, rat, dog, pig, non-human primate and human. We offer single and multiplex chromogenic and immunofluorescent IHC staining for all of your protein localization and co-localization studies.

HSRL has the ability to perform traditional manual staining, and also employs automation using the Biocare Intellipath FLX IHC Stainer. The Biocare stainer is an excellent system for research applications and efficiently facilitates large batch slide runs. Automation allows for increased throughput and consistent results for larger studies.

Our highly trained team of scientists can assist with your research goals by providing input regarding which IHC markers will be most effective for your studies. We are proficient with IHC method development and will optimize the antibodies that fit your research needs. Additionally, we are happy to consult with you regarding recommendations for tissue collection and fixation procedures that will lead to the best possible staining results.

Morphometry / Image Analysis

Morphometric Analysis

Morphometric image analysis of stained slides is a powerful tool that can add value to your drug development studies. Here at HSRL, we perform image analysis using high-quality scanned images.

HSRL employs Visiopharm’s Biotopix™ solution to extract quantifiable data from your stained slides. Morphometry can be used for the following common requests, as well as many more: linear and area measurements, counting positive cells and quantification of positive staining of proteins in cells and tissues. Here are just a few examples of the kinds of morphometric analyses that HSRL can provide for you:

  • CD31 quantification for angiogenesis studies
  • Ki67 or BRDU cell counts for proliferation studies
  • Apoptosis/programmed cell death
  • Immunophenotyping of xenografts, single or multiple immune cell markers
  • Stem cell identification and biodistribution
  • GFAP and Beta-amyloid quantification for Alzheimer’s studies
  • PAS-positive goblet cell counts and area in eyes
  • Insulin and glucagon quantification for diabetes studies
  • Wound healing/re-epithelization in skin
  • Fibrosis quantification in muscle and liver
  • Prussian Blue stain for iron quantification in liver

Data can be provided in Microsoft Excel format or in graph form. We can also provide statistical analyses and histopathological interpretation of morphometry data.

Whole Slide Scanning

Whole Slide Scanning

Converting glass microscope slides to high resolution digital images allows clients to view the slides we prepared from any computer with ease.  Digital slides can be viewed online via HSRL’s image management software, or we can provide the images to you electronically.

Our Hamamatsu Nanozoomer S60 high-capacity whole slide scanner can scan slides at 20X and 40X magnifications. Brightfield and multiplex fluorescence scanning capabilities (up to 6 channels) are available and we can scan both single (1x3) and double-width (2x3) slides. These images can be accessed at your convenience with our image-sharing service, which is very convenient for pathology discussions and internal review, without the need to ship the slides.

COMMON QUESTIONS

Can HSRL pick up our study?
Yes- HSRL has a staff courier and can pick up your study no matter the size or location.
How do I fix specimens for immunostaining?
Place your tissue(s) in 10% neutral buffered formalin for between 24 and 48 hours, then place in 70% ethyl alcohol.
Have you ever been audited by the FDA?
Yes, we have been audited four times by the FDA.  The auditors found no objectionable conditions and no Form 483 was prepared.
